چکیده
Staphylococcus aureus is a leading cause of nosocomial and community-associated infection worldwide; however, there no licensed vaccine available. S. initiates via the mucosa; therefore, mucosal likely to be promising approach against infection. Lactobacilli, non-pathogenic bacterium, has gained increasing interest as delivery vehicle. Hence, we attempted develop an oral based on lactobacilli cushion stress drug resistance needs. In this study, designed, constructed, evaluated recombinant Lactobacillus strains synthesizing nontoxic mutated ?-hemolysins (HlaH35L). The results from animal clinical trials showed that can persist for at least 72 h stably express heterologous protein in vivo. Recombinant L. plantarum WXD234 (pNZ8148-Hla) could induce robust immunity GALT, evidenced by significant increase IgA IL-17 production strong proliferation T-lymphocytes derived Peyer’s patches. conferred up 83% protection pulmonary significantly reduced abscess size skin model. Of particular sharp reduction protective effect offered vaccination ?? T cell-deficient or IL-17-deficient mice. conclusion, first time, genetically engineered induced superior immunity, which was associated with high infections caused aureus. Taken together, our findings suggest great potential system provide experimental data development vaccines

Vaccine protection against Staphylococcus aureus pneumonia
Staphylococcus aureus pneumonia causes significant mortality in hospitalized or healthy individuals, and recent increases in morbidity are attributed to the rapid spread of methicillin-resistant S. aureus (MRSA) strains, which are often not susceptible to antibiotic therapy. Alpha-hemolysin (Hla), a secreted pore-forming toxin, is an essential virulence factor of MRSA in a mouse model of S. aur...

INTRODUCTION Despite aggressive medical and surgical management, the resolution of skin and skin structure infections is often difficult due to insufficient host response, reduced drug penetration, and a high prevalence of resistance organisms such as methicillin-resistant Staphylococcus aureus (MRSA). As a result of these factors, conventional management often consists of prolonged broad-spect...

متن کاملCutaneous Immune Defenses Against Staphylococcus aureus Infections
Staphylococcus aureus (S. aureus) is a virulent bacterium that abundantly colonizes inflammatory skin diseases. Since S. aureus infections occur in an impaired skin barrier, it is important to understand the protective mechanism through cutaneous immune responses against S. aureus infections and the interaction with Staphylococcal virulence factors. In this review, we summarize not only the pat...
